You're certainly not going to be able to create that effect without plugins unless you mean plugins that don't come with AE by default. AE CS3 has the ability to create strokes along paths. This is the first step to creating that effect. Develop your final vector path using Illustrator or AE copy it into AE CS3 onto a Black Solid. Animate the stroke offset so that the line grows with time. Now if you don't have AE CS3 then AE 7 can do it with one additional step. Under the effects tab go to Generate\Stroke and you'll get the same options. Adjust the settings for the stroke itself including brush size and hardness. A low hardness will give you soft edges that'll make the stroke seem to glow. Multiply the Black Solid layer with differing opacities, hardnesses and colour schemes to achieve your final animated stroke. Now comes the tough part. Create a new instance of the black solid and apply Particle Playground from the Effect tab\Simulation. Map the emitter position to the starting point of the stroke and animated it along the timeline so that the emitter follows the 'head' of your stroke as it grows. Play with the setting to create the right kind of particle effect around the stroke (I'm at work or I'd go into the details of that too). However, all this is just so much easier if you just use Trapcode 3D Stroke and Trapcode Particular but some people just want to learn the hard way.
